The detailed mechanism of each step in the formation of acrylamide may depend on the species involved (i.e., the nature of the carbonyl compound) and on conditions of temperature, water content, and pH. The mechanism has been studied in some detail (21, 22, 27, 28). The initial Maillard intermediate N-(D-glucos-1-yl)-L-asparagine, formed from the addition of asparagine to glucose prior to dehydration to the Schiff base, has been shown to release signi cant quantities of acrylamide upon heating at 180 C (21). In simple model systems containing only asparagine and a reducing sugar, the Schiff base of asparagine is probably a major precursor of acrylamide. It has been suggested that the hydroxyl group in the -position to the nitrogen atom will favor decomposition of the Schiff base (28, 29). However, the Maillard reaction produces other reactive carbonyls (Fig. 2.1.2) all of which are capable of forming Schiff bases with asparagine and thus producing acrylamide. Glyoxal, methylglyoxal, and butanedione have all been isolated from asparagine glucose model systems (30, 31). In heated foods, other free amino acids will react with sugars thus increasing the pool of reactive carbonyl intermediates; consequently the asparagine sugar Schiff base may assume less importance as the route to acrylamide. The effectiveness of different carbohydrate moieties in forming acrylamide has been investigated by a number of research groups. Zyzak et al. (16) investigated the ability of different carbonyls to generate acrylamide in a potato snack model system and found that a variety of carbohydrate sources could

generate acrylamide from asparagine, including glucose, 2-deoxyglucose, ribose, glyceraldehyde, glyoxal, and decanal. Schieberle et al. (27) heated equimolar mixtures of asparagine and ve different monosaccharides and two disaccharides at 170 C for 30 min in closed glass vials in the presence of 10% water. Yields of acrylamide ranged from 0.8 to 1.3 mol%, with glucose being the most effective. It is interesting to note that sucrose, a nonreducing sugar, produced almost as much acrylamide as some of the reducing sugars did. This nding suggests that during thermal processing of foods, sucrose may undergo hydrolysis to glucose and fructose. Stadler et al. (15) also noted that glucose, fructose, galactose, lactose, and sucrose all gave comparable yields of acrylamide when heated with asparagine at 180 C. Other carbonyls have been evaluated for their effectiveness in producing acrylamide. Stadler and coworkers (21, 29) compared dicarbonyls and hydroxycarbonyls with glucose, using model systems containing equimolar quantities of carbonyl and asparagine heated, in the presence of water, in sealed tubes at 180 C for 5 min. The highest yields of acrylamide were for hydroxycarbonyls, followed by glucose, with -dicarbonyls, such as butanedione and glyoxal, giving yields much lower than that of glucose. Glyoxal (ethanedial) is a very reactive dicarbonyl and these results contrast markedly with a recent report by Amrein et al. (30), which showed 300-fold higher yields of acrylamide from asparagine glyoxal model systems than from similar models with glucose or fructose. Other recent work has con rmed that glyoxal is very reactive toward asparagine in producing acrylamide (31).

2.1.4 ANALYSIS OF ACRYLAMIDE IN FOODS Products of the Maillard reaction that contribute to color and avor in both model systems and foods have been studied for several decades and, therefore, it is perhaps surprising that it was not until 2002 that acrylamide, a Maillard reaction product, was found in heated foods. However, it has a low molecular weight (71 daltons) and is highly soluble in water. Analytical methods for Maillard reaction products as avor compounds have utilized their volatility while the nonvolatile products studied have had higher molecular weights. Such approaches are not likely to isolate acrylamide. Methods for the analysis of acrylamide in water supplies and in crops had been established well before acrylamide was discovered in heated foods (32, 33). The methods involved bromination to give a dibromo-derivative and separation by gas chromatography. Since acrylamide is very soluble in water, it can be easily separated from the food using aqueous extraction. Water can be used alone or in conjunction with methanol. In some foods the starch gelatinizes in water, making the aqueous layer dif cult to separate from the substrate; the use of aqueous methanol (25%) overcomes this problem (34). Polar organic solvents, such as acetonitrile, ethanol, or propanol have also been used (35). All methods use
